I currently use two L1 Model II towers as my band’s PA. They are connected to a Mackie ProFX16 mixer. I’m thinking about buying a third tower with a T1 and B2 to replace my amp. Would this arrangement work? Here is a link to a youtube video showing my setup:

The distance between L1 #1 and L1 #2 is approx. 20 feet. Assume this is a stage that is 22 feet wide. What is the optimum distance between L1 #2 and L1 #3? What is the minimum distance between #2 & #3 for satisfactory performance?

There are many aspects of sound that are scientific, and some are organic. Placement of the L1®s on the stage is organic. You basically set them up roughly where they should go (the picture above is an example). Then you can fine tune as you get used to the sound.
One of the great things about the L1®s is low drop-off in volume over distance. This means that you have more latitude (flexibility) with respect to where you place them on a stage or in a room.
Coming back to the details of your question…
You want L1® #1 and L1® #2 roughly centred behind the people who are sharing them.
Since you (L1® #3) are not sharing with anyone, just put it behind you. A couple of feet left or right is not going to make much difference although you may have a personal preference.
There is no ideal distance between L1®s that are not shared. If I had to suggest something, I would say that L1®s should be spaced about the same distance apart as the distance between the performers that are using them.
In your suggested positioning, the distance between #3 & #2 is 5’-6’. No problems with this spread? Still good stage sound from all three L1s?

Sounds good to me. Since the sound coming from units #2 and #3 are different, there should not be any adverse interactions.
Thinking back on the video, you may get better sound if you move the #3 out of the corner. Just try it - or maybe just move the B2 closer to the Drums. Having a Bass module in a corner can (in some rooms) give you an exaggerated bass presence.

My four pc rock band runs three L1s (3 T1s, 8 B1s and 2 A1s). Going left (as you face the stage) to right - The bass player/backup singer has a model one and 3 B1s on the end of his side of the stage. My (lead singer/r.guitar) is next with 2 B1s. Then comes the drums. Then the L1/3 B1s that are shared by the drummer/backup singer and the lead guitarist.
The lead guitarist is the only person using a backline amp.
You guys are so close to having the ideal set up - that I’d encourage you to go full tilt and ditch the mixer in favor of 2 or three total T1s. Mix your sound in the air - not your board. I’d also recommend at least one B1 or B2 with each L1.

Just bought a B2 so now both L1s have a B2 bass module. Neither has a ToneMatch engine. Also, just bought an L1 Mod II, B2 and T1. I now own:
3 L1 Mod II
3 B2
1 T1
I see the beauty of mixing in the air rather than the Mackie. Another $1K would buy two more T1s. I could probably ditch the Mackie at this point.
