Using Yamaha MG06X With L1 Compact

Can I pair the MG06X the mixer with my L1 to replace my T1 ToneMatch?

If so, what is the best settings for using the two for Karaoke using a laptop with the MTU karaoke program, Hoster?

Sure - on the L1 Compact, there's a 1/8" stereo input you could use.  But if you want to run the cable more than 20 feet, I'd probably do it the way I showed you above because this will give you a balanced connection.
